Researchers at the Indian Institute of Science Education and Research (IISER) Berhampur have developed a nanotechnology-based biosensing platform aimed at enabling faster and more sensitive molecular detection of Turner syndrome, a chromosomal disorder associated with the loss or partial absence of one X chromosome. The research, led by Dr. Parikshit Moitra from the Department of Chemical Sciences, has been published in Advanced Functional Materials.

Turner syndrome can affect growth and reproductive development and is associated with features such as short stature, delayed puberty and primary ovarian insufficiency. It can also be associated with cardiovascular, renal and other health complications. Early identification can therefore help clinicians plan appropriate evaluation and follow-up.

The current standard diagnostic approach includes chromosomal karyotyping, which requires specialised laboratory facilities. The researchers identified the need for faster and potentially more accessible molecular approaches for detecting Turner syndrome-associated genetic signatures. The new platform is being investigated as a possible route towards more rapid genetic screening.

The technology combines an emiss

ive covalent organic framework (COF) with gold nanostars and gene-specific oligonucleotide probes. Rather than detecting the condition through a single conventional marker, the platform is designed to recognise specific DNA signatures associated with Turner syndrome.

According to the research paper, the sensing system was tested for gene-level signatures involving SHOX, ARSE, XIST, and VAMP7. The researchers used a nucleotide-driven self-assembly process in which the two-dimensional COF and three-dimensional gold nanostar structures interact in the presence of target DNA sequences. This molecular recognition produces measurable signals that can be analysed through fluorescence, UV–visible spectroscopy, and photoacoustic scattering.

The study compared the sensor's responses across 19 rare-disease samples and healthy specimens. The reported limits of detection ranged from 5.2 to 61.2 pg/mL, depending on the genetic target. The study also reported diagnostic accuracy of more than 95% for the XIST signature when compared with established reference methods. These results indicate that the platform has potential as a molecular diagnostic technology, although further validation would be necessary before it could be considered for routine clinical testing.

The research involved scientists from IISER Berhampur as well as collaborators from institutions including Bhabha Atomic Research Centre, Nil Ratan Sircar Medical College and Hospital, and the Institute of Post Graduate Medical Education & Research and Seth Sukhlal Karnani Memorial Hospital in Kolkata.

IISER Berhampur said the technology could eventually contribute to the development of rapid or point-of-care genetic testing, particularly in settings where access to specialised diagnostic infrastructure is limited. The institute described the work as an interdisciplinary effort linking chemistry, nanoscience and biomedical diagnostics.

The researchers have also indicated that the underlying sensing strategy may be adaptable to other inherited disorders. However, this remains a potential future application rather than an established clinical use.

The study represents an emerging approach in precision diagnostics, where engineered nanomaterials are combined with molecular recognition mechanisms to identify disease-associated genetic signatures. If the technology undergoes successful clinical validation and regulatory evaluation, such platforms could eventually complement existing genetic testing methods and support earlier identification of selected chromosomal disorders.
